1 Breast and female reproductive tract cancers (gynecologic cancers) cause high mortality among women and are an important social problem. Recent studies of new anticancer compounds used to treat gynecological cancers are mainly based on 2D in vitro assays. Therefore, we propose research on the development of a Cancer-on-a-chip system, in which a culture model of cancer cells will be obtained taking into account their vascularisation and spatial distribution. The main goal of the proposed project is to develop a new methodology of three-dimensional (3D) modeling of gynecological cancers using perfusion microbioanalytical Lab-on-a-chip systems useful to study basic functions of cancer cells and toxicity assays. A new type of multilayered and vascularized cell culture model will be developed to test and optimize microfluidic methods for examination of biochemical processes in cells based on impedance spectroscopy and fluorescence assays. 2 Research concerns the development of advanced three-dimensional cellular models and new microfluidic systems where in vivo conditions can be simulated. To develop Organ-on-a-chip systems (Heart-on-a-chip, Inslet-on-a-chip) we used photolithography, microfabrication, and 3D printing. Another issue is the development of vascularisation in such culture models (vessels-on-a-chip), thanks to which it is possible to obtain the connection of several organs in one microsystem - Body-on-a-chip system. Such systems can be used to study cellular functions, simulating disease states or study the toxicity of selected therapeutic methods.
