The number of patients suffering from peripheral nerve injuries is steadily increasing each year. Injuries resulting from motor vehicle accidents are pointed out as the main cause of peripheral nerve damage. Recent studies have shown that DNA demethylation is a fundamental mechanism for reprogramming the cellular state of mature mammalian neurons to enable axonal outgrowth. The proposed doctoral research is aimed at incorporating enzymes belonging to the TET (Ten-eleven translocation family) protein family into the structure of a biomaterial and then examining the ability of the resulting system to demethylate the nuclear genome of damaged nervous tissue. Ultimately, the project results will help pave the way for novel therapeutic approaches that provide better recovery of lost sensory and motor functions after peripheral nerve injury.
